Development of cell-targeted lipid nanoparticles for selective delivery to T cells

Lipid nanoparticles (LNPs) have emerged as a leading platform for the in vivo delivery of genetic medicines, although achieving precise, cell-specific targeting remains a significant challenge. In this seminar, I will examine current approaches to engineering cell-targeted "stealth" LNPs, emphasizing the role of lipid composition, surface chemistry, and formulation processes in defining their physicochemical properties and in vivo delivery. I will highlight strategies such as ionizable lipid design and ligand conjugation for selective T cell targeting. Particular emphasis will be placed on analytical characterization and structure–activity relationships that enable extrahepatic delivery, improved efficacy, and reduced off-target effects in emerging immunotherapeutic applications.
